Summary By using 15 available mono/nullisomic lines of Sun II back ground, the Heterodera avenae resistance gene in Nelson (from Avena sativa CI 3444) and Panema (from A. sterilis I. 376) were located on monosome XV. Summary Functional markers (FMs) are derived from polymorphic sites within genes causally involved in phenotypic trait variation (Andersen,
J.R. & T. Lübberstedt, 2003. Trends Plant Sci 8: 554–560). FM development requires allele sequences of functionally characterized
genes from which polymorphic, functional motifs affecting plant phenotype can be identified. In maize and other species with
low levels of linkage disequilibrium, association studies have the potential to identify sequence motifs, such as a few nucleotides
or insertions/deletions, affecting trait expression. In one of the pioneering studies, nine sequence motifs in the dwarf8 gene of maize were shown to be associated with variation for flowering time (Thornsberry, J.M., M.M. Goodman, J. Doebley,
S. Kresovich, D. Nielsen & E.S. Buckler, 2001. Nat Genet 28: 286–289). Proof of sequence motif function can be obtained by
comparing isogenic genotypes differing in single sequence motifs. At current, the most appropriate approach for this purpose
in crops is targeting induced local lesions in genomes (TILLING) (McCallum, C.M., L. Comai, E.A. Greene & S. Henikoff, 2000.
Nat Biotechnol 18: 455–457). In central Europe, maize is mainly grown as forage crop, with forage quality as major trait,
which can be determined as proportion of digestible neutral detergent fiber (DNDF). Brown midrib gene knock out mutations have been shown to be beneficial for forage quality but disadvantageous for overall agronomic performance.
Two brown midrib genes (bm1 and bm3) have been shown to be involved in monolignol biosynthesis. These two and additional lignin biosynthesis genes have been
isolated based on sequence homology. Additional candidate genes putatively affecting forage quality have been identified by
expression profiling using, e.g., isogenic bm lines. Furthermore, we identified an association between a polymorphism at the COMT locus and DNDF in a collection of European

Beet cyst nematodes (BCN, Heterodera schachtii), Cercospora beticola, and rhizomania, caused by the beet necrotic yellow vein
virus (BNYVV) and vectored by the soil-borne fungus Polymyxa betae, are the most serious diseases of sugar beet ( Beta vulgaris
subsp. vulgaris). The wild Beta species of section Procumbentes are known to be completely resistant to H. schachtii, C. beticola
and P. betae. Alien monosomic additions (2n=19), plants of cultivated beet (2n=18) carrying different individual chromosomes
of B. procumbens (2n=18) or B. patellaris (2n=36), were tested in greenhouse experiments for resistance to these pathogens.
Gene(s) conferring full resistance to the beet cyst nematode in B. patellaris are located on chromosome 1.1, and the other
tested chromosomes of B. patellaris are not involved in the expression of resistance. Artificial inoculation under greenhouse
conditions, with in vitro produced inoculum of C. beticola and spot-percentage rating of the disease intensity, showed that
the high level of resistance that was observed in the wild species B. procumbens and B. patellaris was not found in any of
the monosomic additions tested. It was suggested that genes on various chromosomes of the wild species are needed to express
full resistance, and that the chromosomes of group 7 of B. patellaris and chromosome 7 of B. procumbens have the largest effect.
The greenhouse tests for resistance to P. betae in B. patellaris derived monosomic additions showed that the addition families
of group 4.1 have a strong partial resistance, while the addition families of group 8.1 appeared to be completely resistant
to the pathogen. Resistance to P. betae in the two wild species as well as in the two resistant addition types did not exclude
infection with BNYVV, but resulted in a considerable reduction of the virus concentration. It was concluded that resistance
to the vector would complement virus resistance, and may provide a more effective and durable control of rhizomania.

Summary
Azolla accessions (section Azolla) from the germplasm collections of the International Rice Research Institute and Washington State University were fingerprinted and classified by enzyme electrophoresis and leaf trichome morphology. A. filiculoides was enzymatically distinctive and also reliably identified by its prominent one-celled trichomes. Neotropical accessions labelled as A. filiculoides proved to be members of other species. Two groups of isolates were designated A. rubra, but those from Japan were identified as A. filiculoides. The A. rubra of Australia-New Zealand was biochemically unique and possessed less protuberant trichomes than A. filiculoides. A. microphylla, A. mexicana, and A. caroliniana were phenetically similar, but a. microphylla was identifiable from the others in the banding patterns of certain enzymes. A. mexicana and A. caroliniana were closely related enzymatically. The two-celled leaf trichomes of these three species were similar in size and shape. 相似文献

Fungal isolations and fungicide efficacy trials were carried out to determine the possible causes of discoloration in oat groats and potential control measures. Alternaria species and Microdochium nivale were isolated from field samples, and Fusarium poae was found in glasshouse tests. Symptoms could be induced reliably in the field through the use of a mist irrigation system. The cultivar ‘Gerald’ was most resistant followed by ‘Jalna’. Cultivar ‘Image’ was most severely affected by discoloration and ‘Millennium’ was of intermediate susceptibility. None of the 28 fungicide‐timing combinations used in the field trials affected the occurrence of discoloured groats. It was concluded that the best strategy for minimizing risk of discoloured groats would be screening new cultivars or breeding material in mist‐irrigated field trials for genetically based resistance. 相似文献

Summary The regular meiotic behaviour of the cultivated oat Avena sativa (2n=6x=42) is genetically controlled. The factors which control the diploid-like meiotic behaviour also restrict the amount of pairing that occurs between alien chromosomes and their homoeologues in A.sativa, and hence increases the difficulties of introducing desirable variation from wild species into the cultivated oat. A genotype of the diploid species A.longiglumis which interferes with the regular meiotic behaviour of A. sativa can be used to induce pairing between alien chromosomes and their corresponding chromosomes in A. sativa. Using this procedure the dominant gene conferring mildew resistance has been transferred from the tetraploid weed species A. barbata into the cultivated oat. 相似文献

Downy mildew resistance originating from Allium roylei Stearn provides a complete resistance to onions and is based on one, dominant gene. Since A. roylei can successfully be hybridized with onion (A. cepa L.), a breeding scheme aimed at the introgression of this gene was initiated ca. 20 years ago. Several setbacks in this programme were encountered, firstly the identified molecular marker linked to the downy mildew resistance locus became increasingly difficult to use and finally lost its discriminating power and secondly the final step, making homozygous introgression lines (ILs), turned out to be more difficult then was hoped. GISH analysis showed that the chromosomal region harbouring the resistance locus was the only remaining piece of A. roylei in the nuclear background of onion and it also confirmed that this region was located on the distal end of chromosome 3. It was hypothesized that some factor present in the remaining A. roylei region was lethal when homozygously present in an onion genetic background. The identification of an individual with a smaller and more distally located introgression fragment and homozygous ILs in its progeny validated this hypothesis. With the help of these nearly isogenic lines four AFLP® markers closely linked to the resistance gene were identified, which can be used for marker-aided selection. The introduction of downy mildew resistance caused by Peronospora destructor into onion is a significant step forward in the development of environmentally-friendly onion cultivars. 相似文献

The paper compares farmland amenity values generated by a hedonic property value model and a contingent choice model, both estimated from data collected in the Peconic Estuary System of Suffolk County, NY. Understanding the dissipation rates of chemicals in unsaturated and saturated zones of subsurface soils will help determine if reductions of concentrations to acceptable levels will occur. Chemical properties and microbial biomass and activity were determined for the surface (0-15 cm), lower root (50-105 cm), and vadose (175-220 cm) zones in a Huntington silty clay loam (Fluventic Hapludoll) collected from an agricultural field near Piketon, OH. The rates of sorption, mineralization, and transformation (formation of bound residues and metabolites) of atrazine were determined. Microbial activity was estimated from the mineralization of (14)C-benzoate. We observed decreased levels of nutrients (total organic carbon, N, and P) and microbial biomass with depth, while activity as measured with benzoate metabolism was higher in the vadose zone than in either the surface or the root zones. Sorption coefficients (K(f)) declined from 8.17 in the surface to 3.31 in the vadose zone. Sorption was positively correlated with organic C content. Rates of atrazine mineralization and bound residues formation were, respectively, 12-2.3-fold lower in the vadose than in the surface soil. Estimated half-lives of atrazine ranged from 77 to 101 days in the surface soil, but increased to over 900 days in the subsurface soils. The decreased dissipation of atrazine with increasing depth in the profile is the result of decreased microbial activity toward atrazine, measured either as total biomass or as populations of atrazine-degrading microorganisms. The combination of reduced dissipation and low sorption indicates that there is potential for atrazine movement in the subsurface soils. 相似文献
